Mutation details: To define the requirements of the prion protein for conferring susceptibility to scrapie, deletion mutants of the mouse Prnp gene were constructed and used to generate transgenic mice on a background homozygous for a targeted mutation of the endogenous Prnp gene. In this transgenic line, the sequence encoding two of the five copies of the octa repeat (amino acids 68-83) was deleted, in addition to the larger of the two introns.
