Mutation detailsA severely hypomorphic allele was generated after aberrant recombination resulted in the insertion of the entire targeting vector, along with 1.2 kb of plasmid backbone, 304 bp upstream of the endogenous locus. Quantitative RT-PCR showed Procr transcript levels to be less than 5% those of wild-type in a variety of organs (with the exception of the thymus in which levels were ~10% those of wild-type) and ~1% those of wild-type in embryos at E9.5 and E12.5. The vector included 4.4 kb of 5' Procr sequence, a PGK-neo cassette with polyA signal, 3.5 kb of 3' Procr sequence, and a PGK-cda (cytosine deaminase) with polyA signal. (J:80955)
