The authors indicated that short-wave sleep in mice can be measured by EEG activity within the 1-4 Hz range or delta power. Sleep loss results in an increase in delta power whereas excess sleep causes a decrease in delta power. The differences in delta power could be measured as a QTL in 25 strains within the BXD/Ty recombinant inbred line. QTL Dps1 was found on mouse Chromosome 13 with the closest linked markers identified as D13Mit126, D13Mit106 and D13Mit193 with a LOD score of 3.57. The suggestive range was from 37.8 - 53.4 cM.

Dps1 (delta power in slow-wave sleep 1) is a previously identified QTL associated with sleep intensity mapping to mouse Chromosome 13 in BXD recombinant inbred (RI) strains. Short-wave sleep in mice can be measured by EEG activity within the 1-4 Hz rangeor delta power (a measure of sleep intensity). Sleep loss results in an increase in delta power whereas excess sleep causes a decrease in delta power. Peak linkage of Dps1 occurs at D13Mit126 (45 cM; 85.8 Mb) and the QTL interval is flanked by D13Mit231(39 cM; 77.51 Mb) and D13Mit107 (48 cM; 96.69 Mb). This region contains 236 known genes.

In order to narrow the Dps1 region, authors implemented haplotype analysis of 25 BXD recombinant inbred (RI) strains. This strategy localized the QTL to a 13Mb interval between 81.2 Mb and 94.9 Mb containing 44 known genes. Five genes (Tmem161b, EG667132, Cspg2, Msh3, Arsb) located in this interval display non-conservative coding region SNPs and seven genes (2900024O10Rrik, Cetn3, Mef2c, Hapln1, Zcchc9, Homer1, Arsb)display transcript-level differences in the cerebral cortex and/or hypothalamus between sleep and sleep deprived mice.

Homer1 (94.4 Mb) is believed to be a promising candidate gene for Dps1 because the Homer1a splice variant is up-regulated in sleep-deprived mice. Western blot of the cerebral cortex proteins showed increased expression of Homer1 gene product in sleep-deprived C57BL/6J mice, but not in sleep-deprived DBA/2J mice. In addition, a Homer1 promoter region SNP shows correlation to the accumulation of sleep need and sleep deprivation in C57BL/6J, DBA/2J, and BXD RI strains.
