Mutation detailsA cDNA encoding a C-terminally truncated RUNX1 protein was inserted into exon 4 of the endogenous locus via homologous recombination. The encoded protein truncates at residue 446 and thereby retains the trans-activating subdomains and lacks the VWRPY motif. Semi-quantitative RT-PCR analysis of splenic and thymic message showed the levels of mutant transcript in homozygous mutant mice to be similar to the levels of normal transcript in wild-type mice.
