Linkage analysis was performed on 275 129S6/SvEvTac x (129S6/SvEvTac x C57BL/6J)F1 backcross animals to map the herpes resistance locus, Hrl. Parental strain 129S6/SvEvTac is susceptible to infection by HSV-1 whereas parental strain C57BL/6J is resistant. Hrl is tightly linked to Tnfrsf1a on mouse Chromosome 6 (60 cM) and may be a prime candidate gene. Animals homozygous for a mutation in Tnfrsf1a exhibit a high mortality rate when challenged with HSV-1 (LD50=320 PFU) compared to wild type C57BL/6J (LD50>10<5> PFU). Klra8, a gene responsible for resistance to infection by murine cytomegalovirus, maps near Hrl at approximately 63 cM but was excluded as a candidate gene since it does not show linkage to HVS-1-induced mortality. C57BL/6J-derived alleles confer dominant resistance to HSV-1 infection at Hrl.

A locus referred to as Sml (sex modifier locus) was also identified in the 129S6/SvEvTac x (129S6/SvEvTac x C57BL/6J)F1 backcross with no map position given by the authors. Sml affects HSV-1 resistance in female animals.
