192 female animals from a C57BL/6J x (C57BL/6J x DBA/2J)F1 backcross were genotyped at 10 cM - 15 cM intervals to identify QTLs associated with susceptibility to infection by Trypanosome cruzi. Parental strains C57BL/6J and DBA/2J are both susceptible toT.cruzi infection with death occurring 15 to 30 days post infection. However, (C57BL/6J x DBA/2J)F1 hybrid animals are resistant and do not succumb to infection by T.cruzi.

Tir1 (trypanosome immune response 1), a locus previously identified by Kemp (1997), was detected in this study. Tir1 maps to 23.5 cM on mouse Chromosome 17 with peak linkage at D17Mit135 (P=0.000018). The QTL range of Tir1 spans 16 cM - 22.3 cM flanked by markers D17Mit198 and D17Mit168, respectively. C57BL/6J-derived alleles appear to confer additively inherited susceptibility to T.cruzi infection at Tir1.

Tir2 (also identified by Kemp) reached suggestive significance in this study with peak linkage at D5Mit41 on mouse Chromosome 5 (P=0.00059). The QTL range of Tir2 spans 45 cM (D5Mit20) to 52 cM (D5Mit25).C57BL/6J-derived alleles appear to confer additively inherited susceptibility to T.cruzi infection at Tir2.

A suggestive locus at 71 cM on mouse Chromosome 13 was also detected with P=0.00087 at D13Mit35.
