Mutation detailsA floxed neomycin resistance cassette in reverse orientation was placed at the 3' end of intron 3. An additional loxP site was inserted just downstream of exon 4. In the resulting ES cells, exon 4, which encodes 51 amino acid residues of the conserved catalytic domain, is flanked by loxP sites. An AdenoCre construct was used to remove the neomycin selection cassette and exon 4. Corresponding protein was detected by Western blot analysis. These mutant ES cells, prior to cre recombination, were also used in the generation of Sirt1tm3Fwa mice. (J:85542)
