Mutation details: EGFP-1 was inserted, beginning with its ATG start codon, "in frame" after the first 7 codons of the gene followed (in antisense orientation) by HPRT driven by the mouse PGK promotor. This insertion replaced the rest of the gene and 560 bp of the 3' untranslated region. Homozygous mutant mice failed to express target gene mRNA but did express EGFP mRNA.
