Mutation detailsHuman APP sequence encoding amino acids 591 through 695 was adjoined to the human NEFL regulatory sequences as well as the first exon. Northern blot analysis showed expression of the amyloid derived transcripts, approximately 900 nt long, at levels similar to the endogenous App in the brains of transgenic mice. Western blot analysis using an antibody raised against the beta-amyloid peptide showed a pattern of bands that correlates with carboxy terminal fragments identified in human brain.
