The correlation between gene expression and dietary interaction was examined in a population of (BALB/cStCrlfC3H/Nctr x VY/WffC3Hf/Nctr-A(vy)/a)F1 animals. A/a and A/Avy animals were placed on a 70% (calorie restricted) or 100% (non-restricted) diet andliver mRNA levels were assessed for over 18,000 genes using DNA microarrays. Twenty-eight known genes showing statistically significant differential expression between the 70% and 100% calorie diets and A/a and A/Avy genotypes mapped near known diabesity QTLs. These genes may be considered further for candidate genes.

On mouse Chromosome 17, Tbp (8.25 cM) shows differential expression between A/a and A/Avy animals on the unrestricted diet. Diabesity QTL Obq4 (4 cM) maps near candidate gene Tbp. Vegfa (28 cM) shows differential expression between A/a and A/Avy animals on the restricted diet. Diabesity QTL Trbv4c1 (23 cM) maps near candidate gene Vegfa. Ppm1b (50.8 cM) shows differential expression between the restricted and unrestricted diets on the A/Avy background. Diabesity QTL Insq5 (56.7 cM) maps near candidate gene Ppm1b.

Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on a (C57BL/6J x DBA/2J)F1 x C57BL/6J backcross and 25 BXD (B = C57BL/6J, D = DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with TRBV4 CD8+ T cell proliferation during infection with murine gamma-herpes virus. Parental strain DBA/2J is a low responder yielding a low level of TRBV4 proliferation whereas parental strain C57BL/6J is a high responder. Trbv4c1 and Trbv4c2 were identified in the backcross and RI strains an map to mouse Chromosome 17 at 23 cM (LRS = 17.7 at D17Mit136) and mouse Chromosome 6 at 0.5 cM (LRS = 12.2 at D6Mit86), respectively. Trbv41c is located in an interval overlapping the H2 locus and spans 12 cM - 26 cM on mouse Chromosome 17, mapping near Tcte1. Trbv4c2 is located in an interval containing Tcrb and Cd8a and spans approximately 0 - 30 cM on mouse Chromosome 6.
