Mutation details: The transgene consists of a human GFAP promoter, an intervening floxed lacZ cassette, and a mutant SV40 T antigen sequence. The 2.2 kb fragment of human GFAP was used to direct transgene expression in central nervous system cells of the astrocyte lineage.The SV40 sequence contains a deletion for the small t antigen-specific splice signal and a deletion that results in truncation of the large T antigen at amino acid 121. Immunohistochemical staining of brain sections from transgenic animals showed expression of lacZ and not SV40 T121, as expected.
