Mutation detailsA construct containing a lacZ and neo fusion gene, as well as a splice-acceptor site and internal ribosome-entry site (IRES), replaced exon 2. The deleted region encoded the centromere targeting domain comprised of amino acids 29 through 64. Immunofluorescence staining using an antibody raised against the endogenous protein showed a near absence of protein, as weak signals were observed in some cells, in homozygous mutant embryos.
