• Mutation details: Point mutations were introduced by homologous recombination that altered codons 747 and 759 from ones encoding tyrosine to ones encoding phenylalanine. An FRT-flanked neomycin selection cassette that was inserted into an adjacent intron was removed in ES cells by Flp-mediated recombination in ES cells prior to the production of chimeric mice, leaving a single FRT site in place of the selection cassette. Western blot analysis and flow cytometric analysis on platelets derived from homozygous mice confirmed that a mutant protein was expressed on the cell surface. (J:73416)
