235 (C57BL/6J x DBA/2J)F2 animals and 26 BXD recombinant inbred strains were analyzed for microsatellite markers on the X chromosome to identify loci associated with pentylenetetrazol (PTZ) induced seizures. Parental strain C57BL/6J exhibits resistance to PTZ induced seizures whereas DBA/2J exhibits susceptibility. Two significant loci mapped to mouse Chromosome 2, Ptz1a and Ptz1b. Ptz1a spans 28 cM - 50 cM with a peak LOD score of 7.9 near D2Mit61 at approximately 34 cM. Cacnb4 maps near Ptz1a and may be a candidate gene. Ptz1b spans 10 cM - 26 cM with a peak LOD score of 3.2 near D2Mit521 at approximately 15 cM. Cacnb2 maps near Pzt1b and may be a candidate gene. Sequence comparison of regions encompassing Cacnb2 and Cacnb4 did not reveal differences between C57BL/6J and DBA/2J. A suggestive locus was detected on mouse Chromosome 7 which the authors name Ptz2.
