24 BXD (C57BL/6J, D=DBA/2J) recombinant inbred (RI) strains were used to map provisional QTLs associated with sensitivity to quinpirole-induced hypothermia, locomotor activity, and tolerance. Verification of provisionally mapped QTLs was performed on 233 (C57BL/6J x DBA/2J)F2 animals. Parental strain C57BL/6J exhibits higher baseline locomotor activity than parental strain DBA/2J but is more sensitive to suppression of locomotor activity following injection with quinpirole than DBA/2J. DBA/2J shows a greater hypothermic response to quinpirole injection compared to C57BL/6J. A significant QTL affecting sensitivity to quinpirole-induced hypothermia, Hts, maps to mouse Chromosome 9 from 28 cM - 31 cM with a maximum LOD = 4.6 at D9Mit171. Hts also shows suggestive linkage with quinpirole tolerance, baseline locomotor activity, and quinpirole-induced suppression of locomotor activity. Hts maps near dopamine receptor 2 (Drd2), a possible candidate gene. Several suggestive loci were also mapped. A suggestive locus associated with sensitivity and tolerance to quinpirole-induced hypothermia, baseline locomotor activity and quinpirole-induced suppression of locomotor activity mapped to mouse Chromosome 19 from 9 cM - 47 cM with a peak LOD = 2.5 at D19Mit10. Suggestive loci associated with baseline locomotor activity mapped to mouse Chromosome 5 (LOD = 2.1 near D5Mit11 and D5Nds1) and 13 (LOD = 2.7 at D13Mit193 and D13Ncvs42). A suggestive locus for quinpirole-induced suppression of locomotor activity mapped to mouse chromosome 15 (LOD = 2.3 near D15Mit13 and Cyp2d9).
