Mutation detailsHomologous recombination inserted a floxed CMV-hygromycin-thymidine kinase expression cassette downstream of exon 5 and placed a loxP site in intron 3. Cre-mediated recombination in ES cells excised the expression cassette and left loxP sites flanking exons 4 and 5, which encode the localization motifs and the entire catalytic domain of the protein. Notes
Mouse embryo fibroblasts derived from chimeric mice made with Dnmt1tm2Jae/tm4Jae ES cells in which imprinting had been eliminated (J:104860) display a significantly faster growth rate with nearly half (47%) of cells in the G2 or M phase of the cell cycle. These MEF cells become spontaneously immortalized. The chimeric mice experience widespread tumorigenesis.
