Assay type: Immunohistochemistry
MGI Accession ID: MGI:7384958
Gene symbol: Daxx
Gene name: Fas death domain-associated protein
Antibody: Anti-Daxx (M-112; sc-7152)
Detection system: Secondary antibody coupled to Alexa Fluor
Results
Specimen 1 20h female pronucleus: embryonic day 0.5 (more )
Note: early zygote
Structure Level Pattern Image Note
TS1: 1-cell stage embryo Weak Diffuse 1_20h female

Specimen 1 20h male pronucleus: embryonic day 0.5 (more )
Note: early zygote
Structure Level Pattern Image Note
TS1: 1-cell stage embryo Weak Diffuse 1_20h male

Specimen 1 27h female pronucleus: embryonic day 0.5 (more )
Note: late zygote
Structure Level Pattern Image Note
TS1: 1-cell stage embryo Present Regionally restricted 1_27h Noticeable areas of signal localization began to appear in association with the nucleolus precursor bodies (NPBs) in late zygotes. Immunopositive ring-shaped areas were observed around all the NPBs in female pronuclei.

Specimen late zygote (male pronucleus): embryonic day 0.5 (more )
Note: late zygote
Structure Level Pattern Image Note
TS1: 1-cell stage embryo Present Regionally restricted Immunopositive ring-shaped areas were observed around the vast majority of NPBs in male pronuclei. However some male pronuclei (approx. in 25% zygotes) also contained NPBs demonstrating no such labeling.

Specimen 1 46h: embryonic day 1.0 (more )
Note: two-cell stage
Structure Level Pattern Image Note
TS2: 2-cell stage embryo Present Regionally restricted 1_46h Labeling was observed in the heterochromatin rim at the periphery of nucleolus precursor bodies (NPBs) and in heterochromatin zones not associated with the NPBs.

Specimen 1 72h: embryonic day 3.0 (more )
Note: morula stage
Structure Level Pattern Image Note
TS3: compacted morula Present Diffuse 1_72h Diffuse labeling became prevalent again in morula stage embryos, with only a few foci of concentrated signal retained at the periphery of nucleoli.

Specimen 4a: embryonic day 0.5 (more )
Note: late zygote. Double labeled: red - Daxx; green - Atrx (assay MGI:7384973).
Structure Level Pattern Image Note
TS1: 1-cell stage embryo Present Regionally restricted 4a , 4a'' , 4a''' Daxx accumulated in male and female pronuclei. There was no colocalization of Atrx with Daxx immunolabeling at the late zygote stage.

Specimen 4b: embryonic day 1.0 (more )
Note: two cell stage. Double labeled: red - Daxx; green - Atrx (assay MGI:7384973).
Structure Level Pattern Image Note
TS2: 2-cell stage embryo Present Regionally restricted 4b , 4b''' , 4b'' At late two-cell stage, colocalization of Daxx and ATRX was detectable both in the ring-shaped heterochromatin zones associated with the nucleolus precursor body (NPB) periphery and in NPB-unassociated heterochromatin patches. Some zones of Daxx localization did not overlap with ATRX (and vise versa). ATRX was found in 60% Daxx-positive heterochromatin areas associated with NPBs and in 80% Daxx-positive heterochromatin clumps located outside the NPBs.

Specimen 4c: embryonic day 3.0 (more )
Note: morula stage. Double labeled: red - Daxx; green - Atrx (assay MGI:7384973).
Structure Level Pattern Image Note
TS3: compacted morula Present Regionally restricted 4c , 4c'' , 4c''' There was no colocalization of Atrx with Daxx immunolabeling at the morula stage.

Specimen 5a: embryonic day 1.0 (more )
Note: two cell stage. Double labeled: red - Daxx; green - Atrx (assay MGI:7384973).
Structure Level Pattern Image Note
TS2: 2-cell stage embryo Present Regionally restricted 5a'' , 5a Daxx and Atrx colocalization in the nucleus of 2-cell mouse embryos.
