Molecular description: An EGFP fused to a humanized cre (hCre) sequence and preceded by an internal ribosomal entry site (IRES) was inserted into the 3' UTR of the Lyve1 locus by homologous recombination in E14 ES cells. This construct was followed by an FRT-flanked neo selection cassette. Three successfully targeted clones were injected into C57BL/6 blastocysts and chimeras derived from clone 9 were bred to get germline transmission. The neo cassette was deleted in the germline with Flp recombinase. Selective GFP expression in nuclei of Lyve1 +ve cells is detected by antibody staining.
