Molecular description: A mini gene locus including a cDNA encoding a codon improved cre, a translational stop signal, an artificial intron and an SV40 polyadenylation signal , and an frt flanked neo cassette in reverse orientation were inserted into exon 1 via homologous recombination. The neo cassette was then removed by in vivo Flp mediated recombination. Western blot analysis of liver lysates from homozygous mice indicated protein expression at about 10% to 15% that of controls. Cre-reporter activity analysis indicates functional Cre is found in endoderm precursor cells.
