Mutation detailsThe expressed sequence is a chimeric cDNA encoding the tamoxifen-responsive mutant human estrogen receptor ligand-binding domain ERT2 joined, via a 9-aa flexible linker peptide, to mouse T-cell acute lymphocytic leukemia 1 (TAL1/SCL) truncated by 108 bp at its N-terminus. The cDNA was inserted into the p1017 expression vector between the mouse thymocyte-specific, proximal Lck promoter and a human growth hormone minigene beginning within exon 1 and containing splice donor/acceptor sequences and the polyadenylation signal. Thymic mRNA expression was confirmed by RT-PCR analysis. Immunohistochemical analysis of thymic sections with an anti-estrogen receptor antibody demonstrated nuclear translocation of most of the fusion protein following treatment of the mice with OHT, but its retention in the cytoplasm in the absence of OHT.
