    In silico mapping was employed to identify loci associated with resistance to methotrextate (MTX) treatment in the murine air-pouch model of inflammation. Methotrextate significantly reduces inflammation (as measured by reduced leukocyte infiltration) and increases concentration of the anti-inflammatory agent adenosine in C57BL/6J and BALB/cJ, but not in DBA/1J or DBA/2J. A series of 5,268 SNPs spanning the mouse genome at 12 cM intervals was used for mapping analysis.

    Mtxe1 (methotrextate efficacy 1)mapped to 8.7 cM - 21.1 cM and Mtxe2 (methotrextate efficacy 2) mapped to 40 cM - 87 cM on mouse Chromosome 1. These loci are linked to MTX anti-inflammatory efficacy and adenosine upregulation.

    Mtxe3 (methotrextate efficacy 3) mapped to 12.9 cM - 49.8 cM and Mtxe4 (methotrextate efficacy 4) mapped to 60.1 cM - 94 cM on mouse Chromosome 2. These loci are linked to MTX anti-inflammatory efficacy and adenosine upregulation. Dpp4 (35 cM) and Fpgs (21.2 cM) are potential candidate genes for Mtxe3, and Ada (94 cM) is a potential candidate gene for Mtxe4.

    Mtxe5 (methotrextate efficacy 5) mapped to 0 cM - 15.3 cM on mouse Chromosome 3. This locus is linked to MTX anti-inflammatory efficacy.

    Mtxe6 (methotrextate efficacy 6) mapped to 0 cM - 21 cM and Mxte7 (methotrextate efficacy 7) mapped to 21 cM - 53 cM on mouse Chromosome 14. Mtxe6 is linked to MTX anti-inflammatory efficacy while Mtxe7 is linked to adenosine upregulation.

    Mtxe8 (methotrextate efficacy 8) mapped to 0 cM - 17 cM on mouse Chromosome 15. This locus is linked to MTX anti-inflammatory efficacy.

    Mtxe9 (methotrextate efficacy 9) mapped to 0 cM - 25 cM on mouse Chromosome 16. This locus is linked to adenosine upregulation.
