    QTL mapping was performed on 438 F2 animals from an intercross between the acetaminophen nonsusceptible (APN) strain and C3H/HeJ to identify loci associated with caffeine metabolism. The average spacing between markers is 13 cM. Caffeine metabolism QTLs mapped to mouse Chromosome 1 (Cafq1), mouse Chromosome 4 (Cafq2), and mouse Chromosome 9 (Cafq3). Cafq1 is located 85 cM - 95.8 cM on mouse Chromosome 1 with a peak LOD = 9.7 at D1Mit356. No obvious candidate genes are found near Cafq1. Males show a stronger association to the phenotype than females at this locus. Cafq2 is located 31.4 cM - 57 cM on mouse Chromosome 4 with a peak LOD = 3.4 at D4Mit199. Cafq3 is located 28 cM - 48 cM on mouse Chromosome 9 with a peak LOD = 25 at D9Mit248 and D9Mit21. Cyp1a2 is a likely positional candidate gene for Cafq3 based on its known role in caffeine metabolism and differences in protein and mRNA expression of Cyp1a2 between the parental strains.
