Caption Prolonged and more severe neurological deficits after spreading depression in mutant mice. Representative photographs demonstrating the wire grip and neurological deficits after a single spreading depression (SD) in female wild-type (WT) and homozygous Cacna1atm1Maag/Cacna1atm1Maag (R192Q) and Cacna1atm3Maag/Cacna1atm3Maag (S218L) mice. In wire grip test, WT mice showed minimal deficits soon after SD (a), while Cacna1atm3Maag/Cacna1atm3Maag mice continued to fall off the wire for prolonged periods (b). Neurological examination showed leaning (c) and circling (d) in Cacna1atm1Maag/Cacna1atm1Maag and Cacna1atm3Maag/Cacna1atm3Maag mice. SD was evoked in the right hemisphere in all mice. (e) Graphs showing the time course of neurological deficits (only wire grip latency shown) from four representative knockin mice. One or more episodes of transient neurological deterioration (red arrows) were observed after full or partial recovery in 7/7 homozygous female Cacna1atm1Maag/Cacna1atm1Maag knockin, 4/6 homozygous male Cacna1atm1Maag/Cacna1atm1Maag knockin, 3/3 homozygous female Cacna1atm3Maag/Cacna1atm1Maag knockin, and 4/5 heterozygous female Cacna1atm3Maag/Cacna1a+ knockin mice.
