Caption Generation of the Masp2tm1Wjsc allele. (A) Masp2 was disrupted by homologous recombination in embryonic stem (ES) cells using a Masp2 specific targeting construct established in the vector pKO-NTKV 1901. The construct replaces exon 11 (encoding the C-terminal part of the CCP-2 domain), intron 11, and the start of exon 12 (encoding the serine protease domain) with the PGK-Neo cassette, inserting frame-shift mutations and deleting 122 bp of coding sequence, including that for the essential residues for serine protease activity.
