Caption Generation of the Dmxl2tm1a(EUCOMM)Wtsi and Dmxl2tm1c(EUCOMM)Wtsi alleles. The EUCOMM Dmxl2 gene trap allele with conditional potential and the Dmxl2 allele after the action of Cre recombinase. (A) The Dmxl2 gene trap allele (tm1a allele) contains an IRES:LacZ trapping cassette with an acceptor splice site (En2 SA) and a floxed neoR cassette. LoxP sites flank the critical exon 7 and the neoR cassette. FRT sites flank the IRES:LacZ and neoR cassettes (adapted from www.knockoutmouse.org/about/eucomm). The IRES:LacZ trapping cassette and the floxed neoR cassette have been deleted by the FLIP recombinase to generate Dmxl2lox/lox (Dmxl2tm1c(EUCOMM)Wtsi allele) mice. The critical exon 7 was then deleted by the Nestin cre-recombinase to generate both Dmxl2lox/wt and nes-Cre;Dmxl2-/wt. Dashed lines indicate possible RNA splice isoforms. Single-headed orange or blue arrows represent the positions of the PCR primers used to genotype mice.
