Exon 25 is flanked on the 5' and 3' ends by LoxP sites in combination with an FRT flanked neomycin selection marker (Top). The neomycin cassette was removed in a breeding cross of the heterozygous mutant animals with a flippase-expressing animal, leaving behind a single FRT site (Middle). In the presence of Cre recombinase exon 25, the FRT site and one LoxP site are excised leaving behind a single LoxP site (Bottom). | ||||
