• human CD2 is expressed by T cells at various stages of development in the thymus
• a fraction of gamma/delta T cells in thymus, spleen, and gut epithelium express human CD2; a small number of double negative (DN) stage 1 thymocytes (earliest T-cell progenitors) display human CD2 expression
• Background Sensitivity: bi-allelic activation of Tcrb-V8.2 is observed prior to allelic exclusion, with entire double negative stage 2/stage 3 compartment expressing the human CD2 marker, compared to half this expression level in heterozygous mice or B6/SJL F1 mice; this is independent of D-J and V-DJ rearrangement
• SP and DP (single- and double-positive) thymocytes from homozygous and heterozygous mice show a constant level of expression, indicating after V-DJ rearrangement, expression is monoallelic
