• there is dramatic change in CD4 vs CD8 expression profile compared to wild-type
• a subpopulation of CD4 SP thymocytes show surface expression similar to double positive (DP) thymocytes (except lacking CD8 expression)
• intraepithelial lymphocytes (IEL) lose CD8a expression on TCR gamma/delta lineage cells whereas Cc8atm1.1Litt cells have normal CD8a levels
• percentage is decreased compared to controls (from 84% to 65%)
• in thymic lobes after 3 days in culture, ratio of CD4 single positive cells to double positive cells is much higher than in cultured heterozygous organs; percentage of immature CD8 SP thymocytes is seen
• "CD8-negative" SP cells emerge to a significant extent after 7 days of culture, coincident with development of double positive cells
• increased CD4 single positive (SP) cell number from 11% in controls to 29%
• percentage (1.8%) and absolute numbers of CD8+ SP cells (1.9 x106) in thymus of mutants are decreased compared to controls (4.5%, 2.8 x x106 respectively)
• CD8+ T cell numbers in periphery (lymph nodes and spleen) are decreased vs controls
