• in vivo and in vitro analysis of cell growth in mutant embryos show that the proliferative capacity of mutant embryos is impaired by E6.5
• at E6.5, less severely affected embryos are half the normal size
• at E6.5, mutant embryos are morphologically abnormal
• more severely affected embryos resemble egg cylinders
• expression analysis of brachyury, a marker of embryonic mesoderm, confirmed that mutant embryos fail to make mesoderm
• at E6.5, less severely affected embryos show epiblast, no primitive streak and a proamniotic cavity
• at E6.5, no clear boundary between the embryonic and extraembryonic tissues is apparent
• at E6.5, less severely affected embryos show a very thin parietal endoderm
• at E6.5, the extraembryonic region appears disorganized with no compact layer of trophoblast cells and an abundance of giant cells
• Mash2 expression analysis indicates that no diploid trophoblast cells are present in the mutant extraembryonic region
