• RNA polymerase II binding to the Il10 promoter and Il10transcription are significantly (~3-fold) higher than in control animals

• in mutants backcrossed 10 times, thioglycollate-elicited peritoneal macrophages stimulated with LPS show much higher levels of Il10 production than wild-type macrophages after 6 hours
• Il12 p70 synthesis is reduced compared to controls
• in mutants backcrossed 5 times, after LPS treatment, serum levels of Il12 p70 and IFNgamma are reduced, while TNFalpha levels are slightly elevated compared to controls: IFNgamma levels are ~17% of wild-type levels

• 24 hours post-infection, serum levels of Il12 p70 and IFNgamma are reduced compared to controls, while TNFa levels are slightly elevated
• mice infected i.p. with sublethal (to wild-type) dose of L. monocytogenes, mice dies within 8 days of inoculation while wild-type do not die; at higher bacterial doses, mutants show even greater susceptibility and die more rapidly
