Mutation detailsCRISPR-targeting inserted DNA sequences encoded a conditional by inversion (COIN) module, containing a 3'-splice site which allows the expression of the last exon of the gene fused to a histidin-biotin-histidin-tripleHA tag. The COIN allele is inserted in an orientation opposite to the genes direction of transcription. Activation by cre recombinase inverts the COIN module, resulting in splicing of the pre-mRNA to the tagged copy of the exon. (J:82809)
