Mutation details: The following elements were inserted into an intron: the CAG promoter, a lox66 site, an FRT site flanked neomycin resistance gene cassette, inverted sequence coding for EGFP reporter, T2A self-cleaving peptide and HPV16 gene E7 (non-oncogenic antigen), SV40 poly(A) signal sequence, and an inverted lox71 site. The neo cassette was removed through subsequent Flp-mediated recombination. Only after Cre-mediated recombination will the EGFP-T2A-HPV16E7 sequence be flipped and expressed.
