• Mutation details: To generate the targeting vector, 4.6 kb SalI/EcoRV fragment and 7.5 kb XhoI fragments of a Thap1 genomic clone were subcloned into pBluescript which was used as a backbone for the generation of the targeting vector. A loxP-flanked neostop cassette (3.2 kb) was then cloned into Intron 1 of Thap1 (420 bp downstream of Exon 1) and a third loxP site was cloned upstream of the promoter. Correct excision of the loxP embedded sequences was confirmed by HindIII digestion of the vectors harvested from transformed cre/E.coli. (J:94077)
