• Mutation details: A targeting construct inserted an mCherry red fluorescent protein sequence and a loxP-flanked neomycin (neo) resistance sequence-STOP cassette in the 3' UTR of the X-linked methyl CpG binding protein 2 (Mecp2) gene. The endogenous STOP codon was replaced by a 22bp-linker sequence (Lys-Lys-Asp-Pro-Pro-Val-Ala-Thr). Cre-mediated recombination removed remove the floxed-neo-STOP cassette. The targeted allele expresses a chimeric protein with the mCherry peptide linked to the C-terminus of both splicing isoforms of the endogenous gene. (J:82809)
