• Mutation details: The targeting vector was created using the Rosa26 TARGATT site-specific PhiC31 integrase-mediated cassette exchange to contain a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid (CAG) promoter, a loxP-flanked STOP cassette upstream of the Kruppel-like transcription factor 2, lung (Klf2) cDNA sequence, carrying a point mutation which changed lysine to arginine at amino acid position 121 (K121R), fused to an amino terminal HA tag sequence, followed by rabbit beta-globin intron and polyA sequences and attP/B sites, inserted into the Gt(ROSA)26Sor locus on chromosome 6. The construct was microinjected, along with the PhiC31 integrase mRNA, into the pronuclei of zygotes collected from TARGATT Rosa26-attP3 mice on an unknown genetic background. (J:101977)
