Mutation details: This RC::FLTG dual recombinase reporter construct contains a frt-flanked STOP cassette followed by a loxP-flanked NLS beta-gal::STOP cassette in front of ChR2-tdTomato. The ChR2-tdTomato contains a p2A self-cleaving peptide sequence between ChR2 and tdTomato, which leads to the expression of ChR2 and tdTomato as two separate proteins. This allele allows intersectional genetic optogenetic stimulation of different cell subpopulations defined by the overlap of FlipO and cre recombinase expression and can simultaneously be used for intersectional fluorescent labelling of cells since tdTomato is not bound to the membrane.
