Mutation detailsA mini-gene comprising wild type exons 1620 and flanked by loxP sites was inserted into the endogenous intron 1516 of the gene followed by an frt-flanked neomycin selection cassette. A point mutation (TAC/TTC; aa, T > P) was engineered in exon 16 of the endogenous gene. The neomycin cassette was removed via FLP-mediated recombination in the germline. This allele expresses wild-type protein prior to cre-mediated recombination. Upon removal of the mini-gene, a mutant protein is expressed which contains the exon 16 mutation. (J:234984)
