Mutation details: The following elements were inserted into an intron: an adenovirus splice acceptor site, a conditional STOP cassette (loxP site flanked cassette consisting of PGK promoter, neomycin resistance gene, PGK and 3x SV40 poly(A) signal sequences), followed by the Igk signal peptide sequence, anti-Otx2 Igk-V/Igh-V single chain variable fragment (scfv) antibody cDNA with 6x Myc- and His-tag sequence in-frame, P2A self-cleaving sequence, the EGFP reporter gene, and SV40 poly(A) signal sequence. This allele only expresses the antibody and EGFP after Cre-mediated removal of the STOP cassette.
