Mutation detailsThe allele was designed to incorporate a conditional I499V (ATT to GTT) point mutation into exon 12 of the SMAD family member 4 (Smad4) gene. The targeting vector consists of an initial loxP site immediately upstream of the endogenous splice acceptor site of intron 11, coding sequence of endogenous/wildtype exon 12 of Smad4, a SV40 polyA sequence, a neomycin (neo) cassette flanked by self-deletion anchor sites (SDA) and diphtheria toxin A (DTA), which used for negative selection, and a second loxP site. The I499V (ATT to GTT) point mutation in exon 12 is located immediately downstream of the second loxP site. (J:94077)
