Mutation details: CRISPR/Cas9 technology inserted a cassette consisting of a loxP-flanked adenovirus splice-acceptor (adSA), a mEGFP, and three polyA sequences, followed by the H3f3a coding sequence carrying a glycine to arginine mutation at amino acid 34 (p.G34R) into intron 1. GRCm39 has the GGG to AGG mutation resulting in the G to R substitution at amino acid 35. In the absence of cre recombinase, mEGFP is expressed. Upon cre-mediated recombination, the floxed mEGFP can be excised which allows for expression of the mutant H3f3a.
