Mutation details: A vector containing a slice acceptor site, a CAG promoter, a loxP flanked stop gene trap cassette, Map2k1 cDNA with an activating p.K57N missense mutation, an internal ribosome entry site (IRES), and GFP cDNA was inserted into intron 1 via CRISPR/cas9 technology. In the absence of cre-recombination, approximately 5% of all Map2k1 transcripts contain the mutant codon, indicating the gene trap stop is leaky at low levels.
