Mutation details: Using CRISPR/Cas9 technology, the intron 1 of the mouse Gt(ROSA)26Sor locus was targeted to insert LoxP-3x PolyA (Stop)-LoxP (LSL)-NLS-LbCas12a-NLS-HA-P2A-EGFP cassette. LbCas12a is a Lachnospiraceae bacterium (Lb)-derived variant of Cas12a/Cpf1. The LbCas12a gene was flanked by nuclear localization signals (NLSs) to attain better editing efficiency from higher nuclear translocation rates of LbCas12a. A hemagglutinin affinity tag (3xHA) was fused to the C-terminus of LbCas12a, then followed by a 2A self-cleavage peptide (P2A) and subsequent fluorescent marker, enhanced GFP (EGFP).
