• Mutation details: Genomic sequence (230 bp) containing sequences of the 3' end of intron 4 and the 5' end of exon 5 was replaced with a loxP site, the 3' end of a CMV intron, a cDNA coding for exons 5-21, a 4x poly(A) signal sequence, an FRT site flanked neomycin selection marker cassette, a second loxP site, and 5' exon 5 sequence where aspartic acid codon 135 (GAT) was changed to asparagine (p.D135N). The neo cassette was removed through subsequent Flp-mediated recombination. This allele expresses the wildtype transcript and only after Cre-mediated removal of the wildtype cDNA sequence will it express the mutated transcript. The mutation renders the encoded peptide catalytically inactive. (J:348311)
