Mutation details: A loxP site, a copy of the wild type genomic sequence containing exons 3-11 including the stop codon and poly(A) signal sequences, an FRT site flanked neomycin resistance gene cassette and a second loxP site were inserted into intron 2. Codons 79P-120R in endogenous exons 3 and 4 were deleted and IRES sequences and the EGFP reporter gene were inserted into the 3' UTR between stop codon and poly(A) signal. The neo cassette was removed through subsequent Flp-mediated recombination. This allele expresses the wild type peptide. Only after Cre-mediated deletion of wild type exons 3-11 will it express the mutated peptide and the EGFP reporter.
