• Mutation details: The targeting construct inserted a loxP upstream of the cDNA encoding exon 6 through 10 followed by the 3'UTR, six polyadenylation sequences, and loxP site upstream of exon 6. Additionally, a GFP reporter gene was inserted at the end of exon 6 followed by a polyadenylation sequence. Flp-mediated recombination removed the self-excising, FRT-flanked selection cassette. Cre-mediated recombination removed the cDNA encoding exons 6 through 10, allowing expression of the GFP reporter gene. (J:294166)
