Mutation detailsA loxP site was inserted into intron 1 and an FRT site flanked puromycin resistance gene cassette, a second loxP site, and a copy of exon 2 (with CHYSEL (cis-acting hydrolase element) sequence and the EYFP gene inserted in-frame at the 3' end of the CDS) were inserted downstream of the gene. The puro cassette was removed through subsequent Flp-mediated recombination. The floxed endogenous exon 2 was deleted through after Cre-mediated recombination, allowing expression of the reporter peptide in conjunction with the wildtype neurogenin 3 peptide from the exon 2 copy.
