Mutation detailsA DNA construct encoding a fluorescent reporter protein (tdTomato) was inserted into the endogenous first exon (exon 1alpha) of the p16/INK4apart of the gene through homologous recombination. The insertion knocks out expression of the p16/INK4a transcript and does not affect expression of the Arf transcript. A stop codon and poly-A signal were included at the end of the inserted reporter gene. An FRT-flanked neomycin selection cassette was also inserted into the first intron, and is retained in this allele. (J:272509)
