Mutation details: Sequence coding for an HA tag was inserted immediately downstream of the ATG start codon in exon 2, a loxP site flanked neomycin resistance gene cassette was inserted into intron 1 and the following mutations were engineered in exon 2: serine codons 535 (TCG), 347 (TCC), 350 (TCA) and 355 (TCT) and threonine codons 338 (ACA), 341 (ACC), 352 (ACA) and 361 (ACC) were changed to alanine (p.S335A, p.S347A, p.S350A, p.S355A, p.T338A, p.T341A, p.T352A and p.T361A). The neo cassette was removed through subsequent Cre-mediated recombination. The mutations block phosphorylation of the affected residues in the encoded peptide.
