Mutation details: Using standard CRISPR-based homologous recombination techniques in embryonic stem (ES) cells, CAG promoter-Frt-Stop-Frt followed by lox-Stop-lox and Casp3 expression cassette were inserted into the Gt(ROSA)26Sor locus. Chimeras were generated by blastocyst injection and subsequent germline transmission was confirmed by tail PCR. The neo-selection cassette was excised using a phiC31 strain.
