Mutation detailsThe transgene, derived from a clone isolated from an A-strain thymocyte library, contains a 9.6-kb mouse genomic DNA fragment that encompasses the H2-T18a haplotype gene encoding the TLAa-3 antigen. Transgene-derived transcripts and antigen are expressed at high levels in thymus and at lower levels in spleen and lymph nodes, in contrast with the thymus-restricted expression seen in endogenously TL-expressing mouse strains; no expression was detected in bone marrow. The primary transcript, of 2.4 kb, is seen as the minor mRNA species in thymi of strain A (H2-T18a) mice and of C57BL/6 mice congenic for the H2-T18a haplotype. (J:118591)

Two lines of transgenic mice were generated by microinjection of this DNA fragment into fertilized oocytes of strain C3H/HeNCrj, chosen because C3H/He mice do not express H2-T18/TLA antigen from their endogenous H2-T18 gene(s). The line designated Tg.Tlaa-3-1, estimated to carry 7 tandemly-arrayed copies of the transgene, exhibits much higher thymic expression than does line Tg.Tlaa-3-2, with 12 copies. The latter line fails to develop the thymic abnormalities or cancer phenotype of the former.
